Novel fluorescent probe Cy7-APC

Abstract


Stanford researchers have developed a novel fluorescent probe, Cy7-APC, whose characteristics address current limitations in fluorescent technology. This technology provides tandem fluorescent dye conjugates, where one or more Cy-7 dye moieties are bound to an allophycocyanin moiety in an energy transfer relationship. The number ratio of Cy7 dye to allophycocyanin may range from 1 to 30, with the subject tandem dyes providing a bright signal in the 760 to 800 nm range. In addition, probes are provided comprising the tandem dye conjugated with a member of a specific binding pair. This novel fluorescent probe may be used in a variety of ways. It may be conjugated to ligands, receptors, or antibodies for possible use in immunoassays and competitive protein binding assays. It may be used in combination with other fluorescent labels, each having unique spectral characteristics, expanding its usage for cell sorting, diagnostic assays, histology, and fluorescence microscopy. This invention should increase the sensitivity and accuracy of any particular methodology that is troubled by autofluorescence background or Rayleigh and Raman scattering.
